Anastrozole is a potent and selective nonsteroidal aromatase inhibitor. It is used for the treatment of hormone receptor-positive breast cancer in postmenopausal women, either as an adjuvant therapy following initial treatment with tamoxifen or as a first-line treatment option.
Unlike steroidal aromatase inhibitors, anastrozole does not bind to the estrogen receptor. Instead, it binds to and inactivates the aromatase enzyme, which is responsible for the conversion of androgens to estrogens in peripheral tissues. By blocking estrogen synthesis, anastrozole helps reduce the stimulation of estrogen-sensitive breast tumors, thereby slowing or preventing their growth.
Anastrozole is contraindicated in:
- Premenopausal women
- Patients with known hypersensitivity to anastrozole or any of its components
Caution is advised in patients with a history of liver or kidney disease, as well as those with osteoporosis or other conditions that may be affected by changes in estrogen levels.
The recommended dosage of anastrozole is 1 mg taken orally once daily, with or without food. Treatment is usually continued for 5 years or longer, depending on the individual patient’s response and tolerance.
